Among the fluorescent peptide substrates analyzed, the FRET substrate Mca-Pro-Leu-Ala-Nva-Dpa-Ala-Arg-NH₂ displayed the highest specificity constant with MMP-26 (kcat/Km = 3.0 · 10⁴ M⁻¹s⁻¹, pH 7.5, T = 25 °C). The fluorescence was measured at an excitation wavelength of 328 nm and emission wavelength of 393 nm.
